Otto Lutterotti, Ritter von Gazzolis und Langenthal, his full name, was born the son of a tax official and attended grammar schools in Mehrerau and Innsbruck. In 1929, he began studying art history at the Faculty of Philosophy at the University of Innsbruck and joined the student fraternity Austria Innsbruck in the same year. He received his doctorate in 1934.

After completing his studies, Otto von Lutterotti was a trainee at the Graphic Collection of the Neue Pinakothek and the Bavarian National Museum in Munich from 1934 to 1936. In 1936 he became an assistant at the Institute of Art History at the University of Graz and in 1938 at the University of Innsbruck, where he qualified as a professor of modern art history in September 1942. In 1943, he was dismissed as an assistant for political reasons ("due to his Austrian stance").

After the war, Otto von Lutterotti was rehabilitated and appointed full university professor of art history on November 9, 1945. He retired in 1979.
